We are working on MCF7 tumor model on Balb/c AnN.Cg-Foxn1nu/CrlNarl nude mice now. Concerning the estrogen supplement, are there methods other than daily subcutaneous injection that work well? If the mice develops bladder problems after using E2 pellet, are they severe and when does that usually happen? For postmenopausal woman, they might use estrogen paste/ glue as supplement. Is it possible to use such estrogen paste/ glue for nude mice?

There are multiple ways to supply estrogen for MCF-7, but the growth of the tumors (http://altogenlabs.com/xenograft-models/breast-cancer-xenograft/mcf7-xenograft-model/) is going to occur in most cases if the administration does not lead to bladder issues. Slow release pellets are one of the more efficient options, as they have consistent release rates (or so the manufacturers say) as opposed to other routes of dosage.
if you try with food supplemented with estrogens, I dont know if there is a difference, I used E2 in culture cells and it work perfectly, but in vivo the story is different, try using a pellet with E2, at least androgen I read they use that way.
You can use slow release oestrogen pellets from Innovative Research in America. They are a 60 day release pellet (0.72mg/pellet) and are implanted, subcutaneously, on the chest area of the mouse, at least 3 days before cell injection. We implant there as it is easier to scruff the mouse without damaging the pellet and it is less painful for the mouse. Any further questions please contact me.

We saw toxicity starting at about 4-5 weeks post implantation of very low dose pellets. One of the commenters suggested subcutaneous injections every four days of estradiol valerate in peanut oil. We have not tried this yet, but maybe something to consider. Let us know what ends up working for you!
