Hi researchers,
I have been doing cell culture for a few months now and for the first time, I see some contamination in the MCF-7 culture that we were maintaining. I would like to know what kind of contamination this is, is it mycoplasma?
I had washed several times with PBS but it doesn't disappear, so I was convinced its contaminated.

I am unable to open the image. Nevertheless, I would like to suggest that just to rule out the possibility of bacterial contamination you can streak a few microliter of the culture medium containing the cells on to the LB agar plate and incubate the agar plate in the incubator at 37 degrees celsius overnight. Then check for bacterial colonies the next day. Does your culture medium with the cells look turbid ? This is also an indication of contamination.
If you are suspecting mycoplasma contamination you can use the Mycoplasma Detection kit.

Dear Malcolm, Thank you so much for the input. I have attached the images again in jpeg format so if you would like, you can see if you're able to access them. Ok I'll try the method to rule out bacterial contamination.
The culture medium did not look turbid, from the outside, it just looks like our regular culture flasks.
I think if it is not bacterial, then we will have to proceed with the mycoplasma detection.
It seems as fungal contamination and cell morphology was completely changed.. No use even when you wash with PBS twice.
Don't go for splitting with this culture.
Try to revive if you have stock. (Stocks also need to check for contamination)

It might be fungal contamination. If you send a high quality image, l can help you more accurately. Because Some times high confluency of cells may appear this.
