Could anyone help me with choosing a proper concentration and time point to study gene expression changes after treating dopaminergic neurons (differentiated LUHMES cell line) with 6 hydroxydopamine? I got few RT-PCR plates for screening Parkinsons Disease related genes. I would like to find out how overexpression of my gene influence 6 0HDA induced gene expression changes. Different concentration and time points are used in the literature and I can use only one...please share your thoughts - would it be wise to use 100uM and 6 hrs treatment or this concentration is rather to high?
