I want to see the effect of a bacterial protean in THP1 and AGS cell lines. What would be the exact protocol for serum starvation of these cells.
Just to place them in medium without serum.
Depending on the assay PBS could be enough/appropriate (but you must use them soon after putting in PBS).
I usually starve my cells overnight in the medium containing 0.1 to 0.5% BSA but no FBS.
There is no fix rule in this regard what I believe.
People have used 0.5% FBS for 24h.
I used 1% Fetal bovine serum for 16h for primary tendon cells. You may decide, you want to starve enough so that cells are still alive and in good health
For our assays we just starve for 2 to 4 hours with no serum and no additions, just medium
Our protocol is to switch the culture medium (for primary heart cells or cancer cell lines) to the serum-free medium for 24 hr or overnight before test-agent treatment. However, experiments are designed to be terminated 48 hr after drug treatment. Apparently, cells do not grow well in the serum-free medium for a longer period of time.
