DMBA is highly carcinogenic in experimental animals. Large single and multiple doses produce tumors of the skin, breast, and stomach or leukemias regardless of route of administration. Mammary carcinomas appear in a higher proportion and earlier in Sprague Dawley than in Wistar rats exposed to different chemical carcinogen by gastric instillation. Currenrly literature shows that induction is preferred via subcutaneous injections. The incidence of mammary tumor-positive rats is ~70% for subcutaneous and ~65% by gastric instillation. Both techniques are good enough.
