I have insect cell lines, Sf21 and Sf9.
The growth rate of the cell line, Sf9 is decreased from 24 to 50 hours whereas Sf21 growth rate is normal. Could anyone tell me what could be the reason for decrease in growth rate of Sf9 cells?
Currently, we are maintaining the insect cells in a non-humidified incubator at 29°C. Is the decrease in growth rate of Sf9 cells is due to the non-humidified condition?
I don't think humidity could be an issue. I am using an incubator without humidity, I just fill water in the water jacket to maintain the temperature. (Our incubator is old). Have you checked the viability ? You can check that with the dye. Maybe your cells are infected. If you just thawed the cells, then they take time to acclimatize, so you may see slow multiplication. Have you checked changing the media ? Maybe the nutrients are depleted. You have to check one by one, I would say.

I agree with Shivasankari Gomathinayagam about slow multiplication after thawing and depletion of nutrition because of evaporation of media.
