While working on filamentous fungi for the production of cellulase enzyme, more hydrolytic zone was seen in case of CMC plate incubating at 15 degree cel. than 35 degree cel.
But the growth of fungi was more in case of 35 degree cel. than 15 degree cel. It was just reverse.
Why so??? Are there no relationships among hydrolytic enzyme production and fungal growth.? If fungal growth is more at higher temp. then why is there less production of hydrolytic enzyme at higher temp.?

The ideal temperature for growth of fungi 28-35 c , the production of cellulose E. affected by high temperature and may be denatured. So, the detection of these enzyme was decreased at high temperature.Although, during growth procedure of these fungus at high temperature , the EN.PRODUCED AS AT LOW TEMPERATURE BUT THE PROTEIN NATURE OF EN. SUBJECTED TO DESTRUCTION

I think you must choose the temperature at which growth is optimal for you. For ex. Trichoderma produces cellulase, but this fungus has a high growth rate at 30C and it is difficult to see the hallo .Or you can measure the hallo on the reverse of colony, but in this case the enz. index is
The addition of any fungal inhibitors will affected or inhibited the activity of filamentous fungi for production of cellulase E. Most effective method for detection of fungal E. is the cultivation of fungus on recommended liquid medium as SABOURAUDS DEXTROSE BROTH AND INCUBATED AT IDEAL TEMPERATURE AND AFTER RECOMMENDED TIME FOR INCUBATION THE MYCELIAL MAT WERE REMOVED BY FILTRATION AND THE E. WAS MEASURED PHOTOELECTRICALLY AGAINST STANDARD ( THIS IS SUGGESTED SEMI OR QUANTITIVE METHOD
... well yes: what may satisfactory mean? The Explanation of this phenomenon depends on many parameters, at least on these: fungal species - type of medium - amont of substrate in the medium (if any) - diffusion parameters of the enzyme - charge of the enzyme - charge of fungal surface - charge of the substrate (if any) - changes of charges of fungal surface dependent on temperature - activity of the enzyme with respect to pH of the surrounding medium - do you really see diffusion or do you see activity of the enzyme?, differing charge and activity with increasing diffusion distance - interference of your activity of interest with other secreted activities - coagulation of enzymes dependent on the medium concomitantly with lowered diffusion rate - stability of the enzyme - and so on and so forth.
Sincerely: I like your observation very much: Why - seriously, don't you find out? This could be (must not be of course) the start of a really interesting little (and manageable) research project with some biochemical and definitely with ecoligical relevance.
