What is the reason for peak time shift for a single sample in digital chromatography? (for Example: one day in 3 minute , one day in 2 minute , another day in 6 minute and...
Depends. In HPLC (reverse phase) it varies depending on the mobile phase, temperature, column, sensitivity of RT selectivity of the API (molecule of interest)... In my case the chromatographic profile would drift several minutes longer overnight due to moisture absorption by the mobile phase. Live with it!
If your reagents, system, column, and chromatographic conditions are the same then they should not shift. Most probable it could be your mobile phase preparation or you do not give sufficient time to equilibrate the system.
Throughout the analyses, did you check the pressure? Your column's retention time will change if you don't stabilize it. For example, if you begin your analysis with 5% ACN and 95% H2O, you must stabilize your column with 5% ACN and 95% H2O for 2-3 minutes before beginning the analysis.
