I work on lentil.lentil genome size relatively 4.2 Gb. How can I determine if DNA is pure or not after DNA isolation.Thank you very much for your answer
Genomic DNA purity can be assessed by running a spec measurement on the DNA and running a small portion of the DNA on a 1-1.2% agarose gel. Good DNA will give an absorbance ratio 260nm / 280nm = 1.9. If you have good genomic DNA, you will see a band on the gel that runs >= 20000 bp (typically it would run bigger than the largest band on a 1 KB plus ladder). A common contaminant in genomic DNA preps is degraded RNA (which will run at less than 500 bp).
