Hi I usually use citrate buffer as an antigen retrieval but with TUNEL staining it is not working well. My supervisor suggested I try Proteinase K but nobody has used this in brain in the lab only liver. I was wondering what length of time, temperature of incubation and concentration of Proteinase K people use for their antigen retrieval step in brain tissue? Also which buffer do you dilute in ? I have done it in PBS so far.
