Hello everyone,

Currently we are trying to do some pharmacological studies with thalidomide in vitro as well as in mice models.

Regarding in vitro, does someone have expertise using it? I am working with 50 uM dissolved in DMSO. It works well in terms of solubility, but i am concerned about FBS % in the media. I usually lower it to 0.5 - 1% when using these type of hydrophobic compounds. Any opinion on that?

In vivo I've had many problems. I have tried different vehicles to perform IP injections, but in all cases I get a cloudy "solution" (to be honest, it's a suspension).

I have tried Saline + Tween-80 + DMSO in different proportions, heating, etc. Also carboxymethylcellulose (low viscosity) and the result is the same.

We wanted to use quite a high dose (70 mg/kg), so the drug needs to be concentrated enough in order to inject ~50-100 uL per rodent

Any suggestion?

Thanks in advance
