I am using IZON qEV2 columns to isolate exosomes from 100 ml of cancer cell conditioned medium. in NTA analysis I usually see concentration of 2x10^9 particles/ml. But when I proceed with this concentration to isolate total exosomal RNA using Thermofisher total exosomal RNA and protein isolation kit, I dont see detectable amounts (≥ 5 ng/ul) of RNA ( I use Qubit HS RNA quantification method). Do someone has solution for this?

According to my experience 100mL is very low to obtain a good yield of total RNA, you need to collect at least 300-400mL of cell culture to operate RNA isolation very well

thank you for the answer and What is the usual number of particles/ml you get from 300-400ul of cell culture CM. Because number of exosomes released by each cell type may be different. which cells do you use?
