In the same line with the question, have you ever given a thought about the hybridization event between 2 long strain Single DNA sharing a 6-10bp complementary. How strong and how weak is the hybridization binding. I think I need to search for this answer. Do you have your answers?
Please keep in mind that the stringency conditions of your hybridization experiment are very important (i. e., salt concentration and temperature). The lower the salt concentration and the higher the temperature the more stringent your hybridization conditions will be. This is important for the binding specificity of your probe to the target DNA.
