Good afternoon! Does anyone know the meaning of this type of melting curve? I am using 15 ng of cDNA and primers at the concentration of 10 microM.
Thank you to anyone who will answer
No right concentration in probe and primers
My best guess would be lack of amplification. Have you tested your primers with end-point PCR and if so do they give a product?
If you re-try the QPCR and see the same melt curve, you can run your QPCR products on an agarose gel to see if you get bands or not and their size.
Hope this helps!
Thank you very much! That is exactly what I did, I ran on agarose gel those qPCR products comparing them with a good amplification product and I could not see anything, so I had no amplification at all!
I am working with microglia SIMA9 and I will perform a qPCR. If you have already worked with them, can you suggest me one?
