We are working for the development of lateral flow assay by using a polyclonal antibody to the detection of the virus. We got a good intense band of control line on LFA but didn’t get test line band …….. We use carbon conjugation with polyclonal Antibody…..
I assume you check that the antibodies talk to each other & recognize the antigen. You sould try different detergents in the sample buffer. Change the antibody concentration at the test line. Membrane with different pore size also should be tested 135, 140, 180 . Look for aggregates in the conjugate pad.
As Oded Babai already mentioned, you might perform an ELISA independently from your LFA. Often antibodies do not work as expected. Could you deliver more details about the antibody conjugation?
