What is the ideal control sample for samples from tumor biopsy?

I am planning to work on tumor biopsies to detect specific polymorphism. I wonder what is the ideal control sample?

that was helpful, the question is that how can we be sure that even if the biopsy does not have the disease how can we be sure that it still contains the mutation but it does not develop the disease?b
