i am working on effects of gemcitabine in HepG2 cell line and i should use the IC50 of Gemcitabine in the experiments.i found different values in literature and the numbers are highly variable ranging from lower nanomolar to lower micromolar for the same cell line under same conditions.!!!!!!
the IC50 that i got from my experiments are different from loterature and also is variable in each test of mine.....
in each test i got different IC50 and the conditions are the same in all experiments.what should i do?
the source of gemcitabine that i use is powder and the solution of gemcitabine is unstable and each time i should make it...
do you have any suggestion to solve this problem?
Your post probably did not provide enough information about how you store the gemcitabine. You did not indicate the storage temperature or whether you have noticed crystals in your solutions. The following was obtained by doing a 'Net search using the search term gemcitabine stability. "Reconstituted gemcitabine as the hydrochloride salt in the original vials is chemically stable at room temperature for 35 days but may develop crystals when stored at 4 degrees C. The crystals do not redissolve upon warming." My guess is that your culture conditions may be good enough, but your storage conditions for the drug may need to be evaluated. Refrigeration is an almost natural impulse for the storage of chemical solutions to slow down chemical reactions, but crystallization is a physical reaction and gemitabine is chemically stable.

actually i kept my gemcitabine in room temperature for 10 month.and Whenever I want to use drug solution to treat my cells, I make it in time. because in the beginning of my experiments when i made gemcitabine solution and after a week i using it, its toxicity had diminished.So after that, I was preparing a new solution for each cell treatment.
Did you mean that gemcitabine solution also has a 35-day stability at room temperature?

My read is that the 35 days refers to a solution that should be stored at room temperature. Use of the hydrochloride salt should help to render the drug compound water soluble. Ten months may be a bit too long, but, perhaps the vial labels or package insert contain an expiration date.
