Hi all. After growing and inducting the E.coli cultre I extracted the cells by centrifugation but In the brown pellet there are some black spots. I want to know what is these black spots?. I read that It can be death cells, inclusion bodies..

Using a sterile metal tip, carefully scrape a small sample of the "black spots", together with some of the brown pellet (bacteria). Very gently smear the specimen on the center of a microscope slide. Immediately place a thin glass slide over the specimen before it dries up. Under a light microscope and using suitable magnification, carefully examine the specimen. Try to differentiate the "black spots" from the bacterial cells. Hopefully after microscopic examination, you can identify those offending "black spots".
Have you used metal insterts(in german: Öse) to get the colonies from the agar plate. Cheap metal inserts, can incloude a metal corrosion particel as a black spot.
