I'm looking to isolate the total RNAs from yeast and then prepare them for RNA-Seq. I'll be testing the quality of the RNA with a bioanalyzer. But it seems that most options degrade the RNA or are not good at selecting the total RNAs.
My issue with Trizol is that it doesn't seem to be good at isolating small RNAs. I'm worried that one that are kept are biased in some way. Thank you though!
I don't have access to a bead-beater unfortunately, although I've been experimenting with some different lysis techniques such as vortexing with beads, French press, cell cracker...
