* Are you using a 15% Gel? Due to low molecular weight of IL-6 and 10, it is advised to use 15-20% SDS gel.

* If you do not observe any bands in ponceau, increase the loading volume. As IL-10 is less abundant cytokine and may require more loading volume to be detected.
