I'm wondering what's the best plant total RNA extraction kit to use, if the down stream assay will be qPCR? Also this is for corn leaves and roots. I've been using the Spectrum (TM) Plant Total RNA kit (sigma) and it's decent, but I seem to get trace amounts of gDNA in my samples, which is subsequently carrying over to my cDNA synthesis and samples. I've done all the correct things for primer design (i.e. design them so they span an exon gap junction, etc.), but I'm still getting amplification in my no-RT control. I'm guessing clean RNA will help this.

Thanks for the info and advice. I just purchased some DNAse I, so I'll see how it works. I'm using the sigma kit, which has the option of performing a DNAse digestion step on the column (which I usually skip). I'll check out Norgen's products as well.

I like the Qiagen RNA extraction kit for plants. You can also purchase the Qiagen DNAseI enzyme which you can add to the column before before purifying the RNA from the gDNA.
