My project is comparing Serum culturing for 30 and 60 minutes and serum free culturing for 30 and 60 minutes. Therefore I am comparing all 4 variable with 5 ratios for each treatment. What would be the best Statistical test to use? Many thanks
So I worked out the ratio by dividing the 488-GFP value by Dapi for each treatment.
These are the values I have now:
Serum 30 minutes:
0.443709329
0.429182033
0.202280257
0.388070558
0.282401748
Serum 60 minutes:
0.731403202
0.722250628
0.544474153
0.438081195
0.323643202
Serum Free 30 minutes:
0.890455073
1.023145239
1.02898959
1.067835697
1.064191848
Serum free 60 minutes:
0.84629994
0.95062564
0.93624008
0.87223282
0.79874044
So I'm just wanting to compare each variable and figure out whether there is a significant difference between them but im not too sure which test to use.
The statistical analysis depends on the type of your data ( Scale, ordinal, .. ) and the distribution of the data ( Normal distribution or not ), in case of normal distribution you can use ANOVA but in case of non-normal distribution you must use non-parametric tests ( Kruskal Walis test ).
The statistical analysis of data depends on the types of data ,measurement of data and objectives of the data. Objective is very clear i.e. comparision of four treatments ,Wishesso ,if you have four treatments each have five observation of metric in nature(ratio/intrval scale) then apply ANOVA Parametric or if data are in percentage then first use the transformation of data and then apply ANOVA.If data are non metric then use non parametric ANOVA(Kruskal Waliis test)
