What is the best protocol for subcellular fractionation of Gram-positive bacteria?

I need to obtain the outer cell wall and cytosolic fractions of a Gram-positive bacteria, Clostridium sporogenes. What is the best method of doing so, while ensuring purity of both samples? I want to avoid the use of any chemicals that could affect the integrity of the proteins in both fractions, as much as possible.

Hi Ali,

Thank you for you response. You're right, I meant the cytosolic fraction of the bacteria. I have changed it now in the description just to be clear.

Your suggestion of adopting different methods to obtain each component makes a lot of sense. May I ask if you would have any recommended protocols for obtaining the outer cell wall and cell membrane fraction and the cytosolic fraction respestively?
