You probably have to try this protocol mentioned by Gaurang and then test the quality of the RNA you obtain using a device such as the nanodrop machine to test its quality.

You can use ficoll to separate mononuclear cells, wash cells using PBS, add trizol to cell pellet. I can send you the detailed protocol, if you need.
