Generally, HPLC with electro-chemical detection is a simple and robust method for analysing neurotransmitters in brain regions. The brain region is homogenised with PCA/cysteine/internal standard for deproteinisation, spn down and the supernatant can be directly analysed with HPLC-ECD.
Thanks both. Rajesh I had no experience with cyclic voltametric so, anyway thanks once again. On the other hand, I am familiar with HPLC but didn't work with kind a stuff. LI, did you personally done it using HPLCUPLC? If you did would, can you please send my a protocol that you used? I am screening papers on the net and I am finding a very different protocols with a number of different chemicals that are not cheap, so I am in in dilemma about it.
in vivo you can also go with microdilaysis followed by HPLC...just run some aCSF through the region of interest and HPLC the outcome. and HPLC will always be relatively expensive and everyone does it slightly differently so i think you might need to just go with a protocol that looks sensible!
