I have used internode and leaf, but even after increasing the time of sterilization, it is getting contaminated. I only got a minute callus from a leaf disc. What should i do to preserve it? What should be added in the regeneration medium to obtain better callusing?
for preservation i think we need to add some antioxidants to the medium like IAA
I think Cotyledons are good to use as explant. see this protocol, it is very simple . .

Tomato seeds commonly come from rotting tomatoes. This company sells sterile seed hermeticall sealed seed.

You can sterilize seeds from tomato and culture them on MS medium and take the part that you want and put 2,4-D or IBA to increasing the callus.
