Exosomes are extremely stable nanovesicles that are not affected by freezing-thawing and kan be perfectly preserved  at 4oC (short time, reasons see below), -20o and -80oC. One important issue about storage is to ensure that proteins carried on the surface of the exosomes are not destroyed by the action  of matrix metalloproteases (MMPs), especially important if you store them in 4oC, which should be for a short time since beside MMP effect you might get bacterial infection.  A good practice is always to preserve them in a medium/PBS etc. supplemented with a cocktail of metalloprotease inhibitors and if you use them in a few days  to keep them at 4oC, otherwise in -80o. 

Once isolated, exosomes are remarkably stable under various storage conditions. You can keep them at 4 C if you are using them within the next few days.

We store our isolated exosomes at -20 in PBS, both RNA and protein have been isolated from our stocks as long as 1 month following isolation.

 Elizabeth Brandon-Warner- Mam, for isolation of RNA from exosomes ( stored in PBS at -80) , should we directly add lysis buffer to the stored exosomes or again have to pellet down the exosomes. Can we store exosomes in lysis buffer at -80 for RNA isolation ( we use miRVAna miRNA isolation kit)
