I have tried different concentrations of polybrene during experiment but as AML cells are difficult to transfect so could not determine which is the best concentration? To what extent it causes the cell toxicity to death?
Toxicity definitely depends on the cell type - some cells tolerate polybrene better than others. 8ug/ml is a good place to start I think. But yes - AML12 cells are a pain to transduce! Very inefficient
Polybrene is a cationic polymer that can greatly enhance the efficiency of the retroviral or lentiviral infection to the mammalian cells. Usually the range of polybrene conc. 4-8 ug/ml, depending on your cell types. For fibroblast i use 8 microgram/ml but while I use Neuron progenitor cells I don't use Polyrene (it is toxic for NPC).
