What is the best approach to analyze a treatment in a tumor spheroid in terms of fluorescence?

I am looking for the best way to analyze a tumor spheroid in terms of fluorescence and viability. I believe it would be useful to evaluate it by quantifying the area occupied by the fluorochrome, measuring its emitted intensity, and assessing colocalization.
