do you know which procedure should I follow to extract DNA from spider stomach and perform DNA-barcoding? Spiders are preserved under 100% not denaturated ethanol.
there are a couple of Kits designed with stool samples (e.g. DNeasy stool kit - there should be some left at IGB to play around with). But the appropriate chemistry is determined by you downstream analysis. Are you using sanger or next-gen. sequencing?
Another Kit I could recommend would be the Roche High Pure PCR Template Preparation kit.This gave me a very good yield and quality for next-gen seq. Oh, yes and this of course depends on your starting amount of material and the dissection techniques as well. I guess you would try to take the whole spider guts (?) so you would have a lot of spider DNA and a lot less of the spider meal.... . The next big thing to consider - sample homogenization. How do you plan to it? The better the homogenization the more likely to get a better yield.
