I am having trouble obtaining even, longitudinal sections of optic nerve. The nerve looks flat once embedded in the OCT but the sections I obtain are certainly not on an even plane. Any helpful tips or detailed protocols would be so appreciated!

Currently nerves are dissected out fresh, postfixed in 4% PFA for 24 hours, placed in 30% sucrose overnight, then embedded flat in the cryostat directly on chuck (w/OCT) using the Peltier element and heat extractor. Prior to this we had been using traditional OCT molds.

Cutting at 10uM and 4degree angle. -22C cutting temperature.
