I will search for the existence of a protein in 400 samples. I need the best and the cheapest technique that makes the detection of my protein's quantification possible. My budget is limited and this will be just a preliminary data study for an R&D project. The samples including will be body fluid, and my protein has ~105 kDa mass. Western Blot is the unexpensive method in comparison with ELISA. I just thought a semi-quantitative protein arrays (on glass) may be cheaper, but I never used them before. If there is someone who did, I wonder how its trustability and cost compared to Western Blot? (Or any different advice?) Thank you in advance.

This is up to your availiable equipments but the best method for searching a protein in many sample solutions is MALDI-TOF spectometery. It works in low concentration of your protein.

But what about the reliability, isn't necessarily the in vitro validation (basis on antigen-antibody interaction) at the first step to detection the protein existence in body fluid?
In my experience, i think MALDI-TOF is a most important analysis in biochemistry and its too much reliable. But if you have more budget to do another else analysis along with WESTERN Blot, you can do quantative imonohistochemistry staining to detect your protein ( anti gen-anti body reaction) but its expensive. Also, If you know chemical structure of your protein well , you can try ATR machine and its very cheap.
