I'm not entirely sure about the simplest one, however, the best and accurate protocol would be the checkerboard method using broth microdilution. Apart from this, the overlay agar method might be worth trying too. Patch individual bacterial isolate on each side of the agar and let them grow for a few days. Then, overlay with soft agar containing pathogen of interest. If you're looking for antagonistic activity, it would be expected that the zone of inhibition is reduced at the side where the two zones are supposed to come into contact with each other.
