I want to see if isolated bacteria has flagella or not. But I want to do it by using a microscope and without staining. Is there any technique that can help me?
Darkfield microscopy is a simple technique to enhance the contrast of very thin structures without the need for staining. We use this for the imaging of platelet filopodia. This could work for your question too. Regards. Max

If you search PubMed for flagella visualisation there is quite a few papers and chapters which have a lot of useful practical information. Good luck with your project.
