I have this plant extract in which the NCBI paper stated that it is freely soluble with alcohol, ether, and benzene. In water only 28.93 mg/L at 25 deg C (est). And I worried if the alcohol will kill my cell lines because this plant extract will be used for treating some cell lines. What is the best alternative solvent agent for this?

You try to dissolve your plant extract in intermediate polar solvent like methanol. Most of the paper suggests methanol as a good solvent. While you perform your assay using cell-line, keep vehicle control of solvent.

you can try also DMSO which is generally used as solvent for peptodes or probes ( for example Also probes assess cell viability as calcein AM or sytox greenhttps://www.thermofisher.com/it/en/home/brands/molecular-probes/key-molecular-probes-products/live-dead-viability-brand-page.html#prd ) are dissolved in dmso. Of couse you need to dissolve in dmso your plant extract at the massimun concentration and dilute a lot it when you treath the cell lines ti reduce the dmso concentration lower that 0.5-0.1% . As Nutan suggest keep the control with solvent to be sure that the final concentration that you use do not affect your specific cell line.

Hi Idbati - agree with answers above, but are you dissolving a pre-prepared plant extract or creating a plant extract (using ethanol or ether etc)? If the latter then stick with ethanol. DMSO will pretty much dissolve everything - try for 100mg/L or more but either way treat it as a 1000X stock so your final conc. of DMSO/solvent is 0.1% (I commonly use this conc. of DMSO and most cell lines tolerate it - all data is then normalised to 'DMSO only' treated cells alone). Ideally you should try varied % solvents on your cells to test for toxicity. Seed your cells the day before adding the extract. Dilute extract in media to 10X conc (1% DMSO) before finally adding to cells (eg 10uL of this to 90uL cells/media in a 96-well). Do a dose response, don't just add the same dose that you read in a paper.
