with this preparation, you can maintain at least for a while your tissue in the most physiologival conditions to avoid cell death and membranes disruption. The best is always to cool your tissue in a pH controled medium.

Dissection will be made on your filter paper usually wet with PBS. As mentioned, your preparation should be maintained cool and to do so you can use ice covered with a petri dish cover on which you will place your wet filter paper. We use sucrose for enhancing tissue preservation and quality when performing perfusion after the PFA step we soak the brain in sucrose but we do not use sucrose for fresh tissue dissection.

It is a well known fact that using sucrose in PBS acts as a very effective preservative and fixation medium for brain tissues. Normally, we allow the brain tissue to settle down to the bottom of the container for maximum preservation. Similarly, in cryopreservation and cryosectioning also, sucrose is useful for avoiding cracks and ice-crystal holes in the tissue. Hippocapmpus is a very deep located and sensitive area in the brain. The neurons in hippocampus area serve as a very important indicator for measuring neuroprotection and/or neurodegeneration activities for several kind of studies. For example, we involve with plenty of biochemical findings related within hippocampus region during cognitive studies. Unfortunately, such biochemical findings remain speculative unless supported by a histochemical investigation of the hippocampus area also. Thus, using sucrose on the filter paper during isolating the hippocampus area from the entire brain tissue (over iced surface of course) needs expertise and minimum damage to this sensitive area. Sucrose surface helps in maintaining minimum physical or chemical (lysis or osmotic disbalance) damage to the hippocampus tissue for a standard result of the required study.

this is to prevent ice formation in the tissue and keep the osmolarity. You did not describe what hippocampus related research you are doing. You may not need it. For example if you prepare acute hippocampal slices, you may use Ringer solution as more physiological.
