So, I am trying to expand NK cells from PBMC. It seems simple enough, but I am wondering what the ratio of PBMC: irradiated K562-41BBL-IL-15 should be. I did 0.5x10^6 PBMC: 1.5x10^6 K562 cells, and I am culturing them in medium containing 200 U per ml IL-2. Will this work well?

Thank you for your response, and the paper. It is incredibly helpful! (I might just present it at journal club! :-) )
As far as the culture conditions, I have the PBMCs+K562s in 24-well plates at 37C and 5% CO2. I am not supplying them any cytokine, apart from some IL-2 every alternate day. Today (Day 3 post-co-incubation) I am beginning to see the incipience of little, succulent proliferative clumps. So, so far, so good!
