What happen If we add methanol to supernatant? Will they form two layers ? (Organic and aqueous phase)

if you have a water extraction and you add methanol odd are you will have one phase, since water and methanol are miscible.

Methanol is an organic molecule and it will dissolve organic molecules, while a combination of water and methanol is normally used for the extraction of organic compounds like tannis, saponins, alkaloids, steroids etc, as polarity of solvent matter. Dielectric constant at higher temperature also effect the extraction.
Furthermore, if methanol and water mixture is used, it will remain in the same phase
