I can't directly answer your question on which of the 2 cell lines to use, but the one on what is the difference between these 2 cell lines. STF is described here:
https://www.cellosaurus.org/CVCL_AQ26
and STF3a is a STF cell line transfected or transduced (not clear as no precise description was published) with mouse Wnt3a. Here is the preliminary Cellosaurus entry (I found out about STF3a thanks to oyur question and thus will add this cell line to the Cellosaurus):
ID STF3a
AC CVCL_C6NQ
RX PubMed=19022675;
CC Characteristics: Stably express high levels of murine Wnt3a (PubMed=19022675).
CC Characteristics: Responds to activation of the canonical Wnt signaling pathway by increasing expression of firefly luciferase. Luciferase reporter gene expression is under the control of a promoter with 7 LEF/TCF binding sites (from parent cell line)
CC Transfected with: MGI; MGI:98956; Wnt3a.
CC Transfected with: UniProtKB; P08659; Firefly luciferase.
CC Transfected with: UniProtKB; P00552; Transposon Tn5 neo.
