I'm purifying TB antibody and the columns we are using only seem to have a DBC of about 0.15 mg/ml. This seems very low to me, but I am a protein purification novice. Is this normal?
Concentration of your purified fraction is just useless on its own to estimate purification efficiency (as it does vary with the actual volume of the fraction). If you had litres of this fraction (I'm sure you have not), you would have grams of the protein which is great, do you see what I mean? To have an idea of the efficiency, you absolutely need to have some clue on the starting material: what was its content in terms of protein of interest? If your starting material is poor , the purified fraction will be poor too whatever the efficiency of the purification step. Rather than concentration, try to use amount (total mass of protein of interest) and compare these after and before purification (an efficient purification allows to save the protein of interest and get rid of the rest).
