I would like to prevent a protein from becoming ubiquitinated in order to determine whether ubiquitination is responsible for degradation and more specifically if ubiqutination alters function. Obviously, proteasomal and lysosomal inhibitors will only prevent the degradation after ubiquitination. I have looked at some papers with dominant negative mutants of ubiquitin. Anyone have any experience with any of these constructs?
