I want to express a recombinant protein with a Ubiquitin tag at the N-terminus to induce its degradation via the proteasome. Would one ubiquitin sequence be enough or should I add more than one in a row?

No, it may not work with one ubiquitin sequence.
The minimal signal necessary for proteasome targeting is a chain of at least four ubiquitin molecules. This will help for efficient recognition and degradation of ubiquitylated proteins by the proteasome. The polymerized ubiquitin chain acts as a signal that shuttles the target protein to the proteasome, where the substrate is proteolytically broken down.
