Which medium is better for monoclonal antibody production in hybridoma protocol, DMEM or RPMI-1640? Why?
and what are the component of each media?

Use the medium you use for growing up the myeloma cells. I always use RPMI1640 for SP2 myeloma. Immediately after fusion I supplement my medium with extra pyruvate, glutamine and hybridoma enhancer. These can all be obtained from Sigma. The supplements increase my fusion efficiency by more than 10X.

RPMI contains higher levels of glucose I think.
All the additives can be purchased directly from Sigma.
Enhancers contain growth factors from Th cells that stimulate hybridoma growth.
Check the SIGMA media compositions for differences between the two. Will give all the components in each media.
yes.I checked it but you said that you used RPMI 1640 , and I dont know which one is better than another and why? but I know their composition.
DMEM and RPMI 1640 both will work fine ,you can use conditioned medium from macrophage cell (obtained from Unimmunized mice) as a alternate of commercially available hybridoma enhencer.
