Hi,
I need a good inhibitor for CDC42.
I am new to working with this protein and wonder if anyone knows what inhibits it well.
Thank you!
Chuck

Thank you Hassan. ML141 is on my list. Do you have any experience working with it? Are there better inhibitors than it that aren't cytotoxic?
please check this link, it seems to me very useful

Hi Hassan, did you send me a link about medicinal plants of Latin America? Does that have information about CDC42 inhibitors?
Thank you
ML141 is a valid choice only if you don't care about off-target inhibition of Rac1. Unfortunately, I have no experience with other Cdc42 inhibitors.
Thank you Adam. It is certainly on my list. Does anyone else know of any good inhibitors?
Dear Chuck,
Specificity is always an issue with these kind of inhibitors. Similarly, dominant negative Cdc42 mutants fail to ensure full specificity.
You could start with either approach outlined above but, ultimately, I would recommend validating your observations using shRNA or RNAi.
Do not use CRISPR do avoid Cdc42-dependent cell cycle-related phenotypes.
For validated hairpins, have a look at our recent publication:

Good luck!
Metello
Thank you Metello. I will check that paper out. Do you know which dominant negative/positive mutants are the best to study CDC42?
Hi Charles,
DN: Cdc42 G17N
CA: Cdc42 Q61L
These two are the classical ones.
Best,
Hi Chuck, try ZCL278. I recently published a review about cdc42 inhibitors here (Link below). Let me know what you think. Article Two Small Molecules, ZCL278 and AZA197 Show Promise in Influ...
Just a generic question, what is the average half-life of a typical membrane protein? If you have a review off hand that could be sent to me, that would be greatly appreciated.
