I am looking for the best medium composition for rooting of Phalaenopsis in vitro. It is a lot of publication but what is it warking well? Maybe somebody has a proven way...
I think, rooting of the plant in invitro depends on many factors.
1- consistency of agar
2- selection of growth promoting hormones
3- quantity of hormones, micro and macronutients as well
4- temperature, photoperiods etc.
in my opinion, you must survey literature related to your plant tissue culture, then write down important points, like most used hormones, their concentrations, mostly used micro and macronutrients, photoperiods etc.
let me share, sometimes we use mycorrhyzial microflora invivo to improve plant rooting system, can we apply it in your case. Just an idea
Subculturing into a hormone-free medium for a few weeks and then subculture them into a medium supplemented with 0.5 mg/L IAA and 1.0 mg/L NAA or 1.0 mg/L IAA alone might be helpful!
Of course, there are too many factors to stimulate or inhibit or slow down rooting in vitro!
Phalenopsis rooting well has long roots on medium 1/2 MS with 0.5 mg/l BA + 1 mg/l IBA or only 2 mg/l IBA, but the plants are relativ small in compare to long roots. What we can add to the medium for much more faster growth, bigger leaves...ect?
