I purchased and have been using the gold nanoparticle which was stabilized in citrate buffer.
As far as I know, the citrate buffer is commonly used for capping gold nanoparticle as its own electrostatic feature.
In the middle of the process of functionalization of the gold nanoparticle with oligonucleotide probe in phosphate buffer, there was a problem with regard to its aggregation. What if I use distilled water instead of the buffer?

You should try to increase the PBS concentration in order to increase the loading of the oligonucleotides onto the AuNPs. However, You can try to centrifuge and wash but some aggregation is almost inevitable because citrate gives negative charge and Au is positive charged, so aggregation will occur. you can try to use PVA or an viscous media to prevent the aggregation and you can follow this through zeta-potential analysis.
If it is extreme aggregation, the solution will get coloured and then there’s nothing that you can do to recover at this time.
but you should treat you oligonucleotide before to prevent disulfide-bonds.
I believe that if you use distilled water gold inorganic complexes are going to be produced and it is going to be worse than before because the opposite that is happening now is going to happen, nothing is going to be available for the functionalization process.

The nanoparticles are most likely dispersed in water and have adsorbed citrate ions on their surface. I do not use oligonucleotides, but have some questions. Are the particles stable in phosphate buffer? If the olinucleotide is negatively charged and so are the phosphate molecules, are not they going to compete for the same adsorption sites?
