I would like to do an ELISA to measure the levels of a biomarker in mouse serum. The concentration of the standards ranges from 2000pgs/ml to 31.25pgs/ml. I have the kit from R&D systems but wondering what dilution of serum sample should I use?
the scientific way will be you optimize yourself, taking whole serum and some dilutions. In that way you will optimize your kit also and will decide how to go with your experiment
If you do not know the anticipated level of your targeted biomarker in mouse serum, then I would suggest you to just make an attempt to find it out in your system by taking two rows (8 wellsx2) of micro-wells for it.
Take out two strips of ELISA plate and in the first lane add standards ranges from 2000pgs/ml to 31.25pgs/ml. And in the second lane, add diluted serum ranging from (0; 1:1; 1:5; 1:10; 1:20; 1:50; 1:100) dilutions. Check the OD of your diluted samples. Find out the particular dilution that falls under lower-middle concentrations of standards. Thereafter, dilute all samples accordingly and run you full length ELISA to determine the levels of a biomarker in mouse serum.
