I am working on bioethanol production from terrestrial and salt tolerance yeast strain.While estimating the intracellular glucose and glycerol content I have used yeast strains grown in YPD media from 0 to 6 hours at 30 degree celcius. In order to perform lysis I have used a) water bath based sonicator set at 80 degree for 15 mins b) Probe sonicator for 180 seconds at 30 percent amplitude Since the yeast grows from 0 to 6 hrs, the intracellular glucose and glycerol level are expected to increase with time.However, I have got inconsistency in both the glucose and glycerol levels from 0 to 6 hours upon HPLC based quantitation. Please suggest any alternative method for lysis if necessary. Thanks in advance.
