I have 1 mg of avidin and 100 mg of biotin. How do I prepare the stock solution? I want to study the interaction of these two using electrochemical workstation? Is 1:1 ratio followed?
You can have up to 4 biotin molecules bound to avidin.
So, depending how many molecules you want to bind, you use the molar ratio accordingly.
The concentration you prepare for biotin and avidin will depend on this molar ratio.
For example: 1:4 (avidin:biotin)...the stock solution of biotin should be 4x higher concentrated than avidin, and then you just add equal proportion of volume from each stock to the final sample.
