I am working in cell culture of normal human dermal fibroblasts and I face a new challenge :
detachment and death of the cells are occuring for almost all of my different patients in the same time and the culture medium is yellow but clear.
What do you think it can be ? Mycoplasma (even if I use MycoZap in cell culture medium to prevent it) ? or other else ?
I attach pictures to evaluate the damages.
You can check for mycoplasma using DAPI stain.
You can prevent fungal using amphotericin b.
It does look most like fungal from this point of view. However, not 100%. Either way you will have to dispose of your cultures.
The other option that comes to mind is nutrition. The media turns yellow from pH change. This can also happen if you media is not being changed regularly enough (and would also explain why the media is still clear). How often do you change media? have you considered changing it more frequently.

Unfortunately, I do not observe any visible fungal in my flasks ...
I am using amphotericin b + penicillin/streptomycin + mycozap as antibiotic cocktail in the culture media.
I change the media every two days. The culture media turn into yellow when cell detachment is observed. I will check the pH of cell supernatant.
